Secreted Protein Methods

Cell Culture Harvest

Collect cell culture supernatants from expression systems (CHO, HEK293, E. coli, SF21, SF9, T.ni etc.)
Evaluate sample (VCD) and cell density

Centrifugation Method

Place filter plate (PN 921547) on top of collection plate (PN 931569-G-1X)
Aspirate and dispense up to 7mL cell culture supernatant to empty filter wells
Set centrifuge settings to RCF and spin at 336 RCF (340 RPM) for 5 minutes
Remove filter plate and collect clarified samples
Confirm complete clarification and ensure absence of visible particulates
Measure protein concentration in clarified samples (Octet, SDS-PAGE, HPLC/FPLC etc.)
Assess biological activity, if applicable

Cell Culture & Purification-Intracellular Protein Method

The Thomson Universal Rapid Clear® 24-Well Filter Plate demonstrated excellent performance across various cell culture systems and protein types. It achieved optical clarity suitable for downstream analysis via the non-binding depth filter matrix for 90-95% recovery from cell lysate.

Gel shows 4-12% BisTris SDS-PAGE in 1x MOPS, 45 min run, stained overnight with InstantBlue. Loaded 5µL of Streptactin XT elution (~240µL total) per lane; 5µL PageRuler Plus ladder for sizing.
